![]() ![]() ![]() This method details how to take TEER readings using an EVOM with chopstick electrode. An Epithelial Voltohmmeter (EVOM) is used to take TEER measurements. EVOM™ Auto produces a low AC current that avoids electrode metal deposits and is specially designed for the non-destructive, high throughput screening of epithelial monolayer confluence in cell cultures. SCOPE OF APPLICATION (LIMITATIONS) Trans-epithelial Electrical Resistance (TEER) can be used as a measure of cell monolayer confluence, health, and integrity. The EVOM™ Auto automates measurements of TEER in epithelial or endothelial monolayers cultured on high throughput screening 96-well plates utilizing WPI's innovative EVOM technology, qualitatively measuring cell monolayer health and quantitatively measuring cell confluence by determining an increase or a plateau in tissue resistance.Īutomated measurement of tissue resistance in cell culture microplates provides the advantages of speed and precision, minimizes the chances of contamination, and ensures the rapid availability of measured resistance data. The specimen holder was placed in the ENDOHM-6 chamber and contacted using two pairs of electrodes connected to the commercial EVOM-2 voltohmmeter, as was also described in Section 2.3, after which TEER measurements were performed. An EndOhm chamber connected to an Epithelial Voltohmmeter (EVOM) may be used to take TEER measurements that are more reproducible than the chopsticks. EVOM was the first instrument designed to perform routine Trans Epithelial Electrical Resistance (TEER) measurement in tissue culture research. Fluorescent recovery in the lower chamber (well) was measured after 15 minutes as stated in Section 2. Solute permeability (b) was assessed by adding 100 l of 1 mg/mL fluoresceinated dextran-70 in Transwell TM inserts.
